PRINCIPLE:

Molecules containing p-electrons or non-bonding electrons (n-electrons) can absorb energy in the form of ultraviolet or visible light to excite these electrons to higher anti-bonding molecular orbitals. UV-Vis Spectrophotometer records the excitation in the form of a spectrum (absorbance or transmittance versus wave length of light)

APPLICATION:

Analysing the position of absorption band of the spectrum and comparing it with known samples, presence of different functional groups and qualitative detection of unknown compounds can be undertaken. UV/Vis spectroscopy is routinely used in analytical chemistry for quantitative determination of different analytes, such as transition metal ions, highly conjugated organic compounds, and biological macromolecules.
