Abstract

Parkinson’s disease is a progressive neurodegenerative disorder associated with aggregation of α-Synuclein (αS), resulting in formation of plaques in neurons described as Lewy bodies. αS is a small, soluble and intrinsically disordered protein that upon gaining amyloidogenic conformation forms rigid cross-β sheet structure with fibril-like morphology via cytotoxic oligomeric intermediates. Metal ions play an important role in provoking generation of oligomeric species that increases the probability of αS amyloid fibrilllation. In last decade, nanoparticles have gained momentum due to their nano-size and large surface to volume ratio. This provides relatively larger surface to interact with biomolecules in biological milieu. The binding of proteins or nucleic acids onto nanoparticle surface is likely to induce conformational rearrangements, thereby anticipated to impede them in folding pathways and affect overall bio-reactivity of nanoparticle. Here, I am proposing to study the fibrillation kinetics, covalent modifications and cytotoxic propensities of αS and conformational changes in RNA in presence of different nanoparticles. This would involve studying the structural characteristics of αS and RNA in presence of several chemically and biologically synthesized nano-formulations.
